Isopropyl-beta-D-thiogalactopyranoside (IPTG) has the following advantages: 1. Molecules that can induce enzyme synthesis, but are not decomposed, are called comfort inducers. Because lactose can induce enzyme synthesis, but it decomposes with it, resulting in many complex kinetic problems, comfort inducers are often used for various experiments. 2. IPTG is not metabolized by bacteria, is a lactose analogue, once it enters the cell, it will produce a long-lasting induction effect, so IPTG induction efficiency is high, often only need a small amount (1mmol/L) to achieve the ideal induction effect. Since IPTG is not metabolized, the expression of exogenous proteins is specifically induced intracellularly. It is not affected by cell metabolism, and the induction effect is continuous and stable. Lactose has a double effect, can be used as an inducer isolactose precursor, but also as a carbon source, in the induction process, very unstable, IPTG because of its excellent stability determines that it is suitable for experimental research. 3. IPTG can be efficiently copied in the absence of lacY gene. 4. Sulfur is used instead of oxygen in the galactosidic bond and loses hydrolytic activity, but the affinity of thiogalactoside and homologous oxolate compounds is the same as that of the enzyme site, and IPTG is not recognized by β-galactosidase, it is a very effective inducer of lac gene clusters.
