Acetohydroxamic acid is used to treat bacterial biliary, kidney stones and liver coma.
Acetohydroxamic acid can chelate with the nickel atom of urease to inactivate urease, thereby preventing urea from being decomposed, restoring normal urine pH, improving the physiological environment of supersaturation of magnesium ammonium phosphate and carbonate-containing apatite, preventing the formation of stones and inhibiting their growth. Acetohydroxamic acid is used to treat urinary tract infections of refractory urease bacteria, infectious urinary tract stones, stones caused by long-term urinary catheters and other diseases.

Acetohydroxamic Acid (AHA), marketed under the brand name Lithostat, is a synthetic medication that acts as a urease inhibitor.
Its main and approved use is as an adjunct therapy for chronic urinary tract infections (UTIs) caused by urea-splitting bacteria (like Proteus species). These bacteria produce the enzyme urease, which breaks down urea into ammonia, making the urine alkaline and promoting the formation of struvite kidney stones.
AHA works by inhibiting the bacterial urease enzyme. This:
Acidifies the urine.
Prevents further stone growth.
Helps dissolve existing stones.
It is used alongside antibiotics and surgical stone removal.
Major Risks: It is contraindicated in pregnancy (teratogenic) and can cause serious side effects like hemolytic anemia and liver toxicity.
Monitoring: Requires regular blood tests.
Status: Its use has declined due to these safety concerns and the effectiveness of modern antibiotics and surgical techniques.
Sometimes used in veterinary medicine for similar conditions.
Has been investigated for other conditions involving urease, like Helicobacter pylori infection, but is not a standard treatment.
