Iron-dextran is an anti-anemia drug. Iron is the main component of hemoglobin and myoglobin. Hemoglobin is the main oxygen carrier in red blood cells. Myoglobin is the site where muscle cells store oxygen for oxygen demand during muscle exercise.
Ferric dextran is a complex of iron and dextran in solution intended for injection. After intramuscular injection, part of the iron is engulfed by macrophages into fibroblasts, and the rest of the iron is slowly removed (about 50% at 72 hours) and transferred to plasma through lymph. Its absorption route is not controlled by the gastrointestinal tract, so the storage of iron is faster and recovers completely. However, hemoglobin still rises at the same rate as oral preparations.
Iron dextran has the advantages of small toxic side effects, stable properties, good solubility and high iron content, and is used as an iron supplement in the pig industry. Clinically, it is often used to determine the cause of chronic blood loss, malnutrition, pregnancy, child development and other iron deficiency anemia.

Dihydrostreptomycin Sesquisulfate Salt — Detailed Description
Chemical Identity & Structure
Dihydrostreptomycin sesquisulfate salt is a semi-synthetic aminoglycoside antibiotic derived from streptomycin via catalytic hydrogenation of the aldehyde group at the C-1 position of the streptose sugar moiety. This reduction converts the reactive aldehyde to a primary alcohol, yielding dihydrostreptomycin, which is then formulated as a sulfate salt.
The term "sesquisulfate" denotes the precise stoichiometric ratio: 1.5 moles of sulfuric acid (H₂SO₄) per mole of dihydrostreptomycin base, resulting in the molecular formula (C₂₁H₄₁N₇O₁₂)₂·3H₂SO₄ with a molecular weight of 1461.42 g/mol. The CAS registry number is 5490-27-7.
Structural Features
The molecule contains three sugar units — streptidine, streptose, and N-methyl-L-glucosamine — linked by glycosidic bonds. These multiple amino and hydroxyl groups are responsible for its high polarity, strong hydrogen-bonding capacity, and excellent aqueous solubility. The sulfate counterions provide ionic stabilization and enhance crystallinity.

Mechanism of Action
Dihydrostreptomycin exerts its antibacterial effect by binding irreversibly to the 30S ribosomal subunit, specifically targeting the 16S rRNA component. This binding disrupts bacterial protein synthesis at two critical stages:
Initiation interference — prevents formation of the 70S initiation complex, blocking the start of translation
Elongation disruption — causes misreading of mRNA codons, leading to incorporation of incorrect amino acids and production of non-functional or truncated proteins
The cumulative effect is bactericidal — bacterial cells cannot produce essential proteins and undergo cell death. This mechanism is concentration-dependent, meaning higher drug concentrations achieve faster and more complete bacterial killing.
Spectrum of Activity
Dihydrostreptomycin sesquisulfate exhibits broad-spectrum bactericidal activity against:
Gram-negative bacteria — Escherichia coli, Klebsiella pneumoniae, Proteus spp., Pseudomonas aeruginosa, Salmonella spp., Pasteurella multocida, Brucella spp.
Gram-positive bacteria — Staphylococcus aureus (some strains), Streptococcus spp., Enterococcus spp.
Mycobacteria — Mycobacterium tuberculosis, Mycobacterium bovis
Other pathogens — Yersinia pestis (plague), Francisella tularensis (tularemia), Leptospira spp.
Uses & Applications
Veterinary Medicine (Primary Commercial Use)
This is the dominant application for dihydrostreptomycin sesquisulfate salt:
Cattle — respiratory infections (shipping fever complex), mastitis (intramammary infusion for dry-cow therapy), leptospirosis, and calf septicemia
Pigs — bacterial enteritis, respiratory diseases, and post-weaning infections
Sheep & Goats — pneumonia, foot rot, and systemic bacterial infections
Horses — leptospirosis, strangles, and other susceptible bacterial infections
Poultry — pullorum disease and fowl cholera (in some markets)
Formulated as injectable solutions (e.g., 25% w/v for IM use), oral powders for water medication, intramammary tubes, and boluses
Cell Culture & Biotechnology Research
Used as a supplement in cell culture media (BioReagent grade, ≥95% HPLC purity) to prevent bacterial contamination without affecting mammalian cell growth
Employed in aminoglycoside uptake studies to investigate bacterial membrane transport mechanisms
Used in mechanosensitive channel research and hair-cell bundle electrophysiology studies
Pharmaceutical Reference Standard & Quality Control
Available as a certified reference material traceable to Ph. Eur. and USP pharmacopoeial standards
Used for method development, validation, and quantitative analysis of dihydrostreptomycin in bulk drugs, veterinary formulations, and residue testing in food products
Agricultural Crop Protection
Applied as an antimicrobial agent in certain crop protection programs to control bacterial plant pathogens
Used in seed treatment formulations in some agricultural markets
Historical Human Medicine (Largely Discontinued)
Previously used for tuberculosis treatment and serious infections (plague, tularemia, brucellosis)
Clinical use in humans has been largely abandoned due to a significantly higher risk of irreversible ototoxicity (cochlear damage leading to permanent deafness) compared to streptomycin, which primarily causes vestibular toxicity
Selling Points
Broad-Spectrum Bactericidal Efficacy
Dihydrostreptomycin sesquisulfate delivers concentration-dependent bactericidal activity against a wide range of Gram-negative, Gram-positive, and mycobacterial pathogens. This versatility makes it a reliable first-line antimicrobial in veterinary practice across multiple species and infection types.
Superior Chemical Stability vs. Streptomycin
A critical advantage over the parent compound streptomycin: dihydrostreptomycin is not inactivated by cysteine, hydroxylamine, or semicarbazide, and is resistant to degradation by heating with alkali. This enhanced chemical stability translates to longer shelf life, more robust handling during manufacturing, and greater formulation flexibility.
Excellent Aqueous Solubility
With solubility exceeding 50 mg/mL in water, it dissolves readily for formulation into injectable solutions, oral drinking-water medications, intramammary infusions, and cell culture media — eliminating the need for complex dissolution aids or co-solvents.
Cost-Effective, Mature API
As a well-established, off-patent molecule with decades of manufacturing history, dihydrostreptomycin sesquisulfate is produced at scale through fermentation of Streptomyces griseus followed by extraction, purification, and catalytic hydrogenation. Multiple global suppliers ensure reliable supply chains and competitive pricing — making it economically attractive for large-volume veterinary formulations.
Multiple Pharmaceutical Grades Available
Suppliers offer the product in veterinary bulk grade, BioReagent grade (≥95% HPLC purity), and certified reference material grade — serving diverse customer segments from bulk veterinary drug manufacturers to research laboratories and regulatory testing agencies.
Pharmacopoeia Recognition & Regulatory Clarity
Listed in major pharmacopoeias (USP, Ph. Eur.) with established monographs, defined acceptance criteria, and standardized assay methods. Acceptable Daily Intake (ADI) values have been set by JECFA/WHO, and Maximum Residue Limits (MRLs) are defined for food-producing animals — providing a clear, well-defined regulatory pathway for product registration in global markets.
Established Combination Therapy Potential
Dihydrostreptomycin is frequently combined with penicillin, ampicillin, or tetracyclines in veterinary formulations to achieve synergistic bactericidal effects and broaden the spectrum of coverage — a well-documented combination strategy that adds value for formulators.
