Closantel sodium is a halogenated salicylanilide anthelmintic (CAS 61438-64-0; commonly supplied as the dihydrate). It is a veterinary-only antiparasitic drug used mainly in sheep and cattle — it is not licensed as a human medicine in most countries. It was originally developed by Janssen Pharmaceutica and is now widely marketed as injections, oral suspensions/drenches, boluses, tablets and pour-ons, often in combination with ivermectin, albendazole or levamisole.

How it works

It acts as a proton (H⁺) ionophore / uncoupler of oxidative phosphorylation: it shuttles protons across the parasite's inner mitochondrial membrane, collapsing the proton gradient and blocking ATP synthesis. The parasite undergoes spastic paralysis and dies. Because it targets energy metabolism rather than nematode neurotransmission, it has no cross-resistance with benzimidazoles, levamisole or macrocyclic lactones. Its activity is strongest against blood-feeding parasites, which take up large amounts of the highly albumin-bound drug from the host's blood.
